DOES BEMITIL
CHANGE GENE EXPRESSION?

The investable question is whether Bemitil engages a reproducible gene-expression program that improves function under heat at tolerated exposure. We are financing the experiment to find out.

01 / WHAT EXISTS

Indirect evidence of RNA involvement

PMID 12227091 ↗ reports a rat hypoxia experiment in which actinomycin D inhibited Bemitil-associated protection of glutathione-related measures. Transcription dependence is a mechanistic lead. It does not identify regulated genes or establish causality for a human heat benefit.

02 / A SECOND SIGNAL

Enzyme induction needs mechanistic follow-up

PMID 12227093 ↗ examines CYP450-related enzyme activity in rat liver and human lymphocytes in vitro. Those activity measurements are not a transcriptome or a clinical gene-expression study. Enzyme induction also raises drug-interaction questions for development.

03 / RELATED ACTOPROTECTOR

Bromantane has gene-expression research

A 2004 Ladasten study reports tyrosine-hydroxylase and DOPA-decarboxylase gene-expression findings in rat brain. This is bromantane-related evidence, with brain-region and timing dependence. It cannot be transferred to Bemitil or treated as human validation. PMID 15500036 ↗.

04 / WHAT WOULD COUNT AS YES

Reproducible biology linked to function

Require a reproducible expression response, confirmation beyond a single profiling assay, functional heat benefit at tolerated exposure and independent replication. Distinguish effects of the drug from heat itself and from cellular injury. A causal mechanism requires additional perturbation and rescue evidence.

05 / WHAT WOULD COUNT AS NO

A platform thesis with a stop condition

By month nine, reject the expression-platform thesis if no reproducible, non-toxic expression response tracks with function. Stop candidate advancement for failed functional replication or unacceptable safety. Keep the claim proportional to the result.

06 / WHY THE UPSIDE IS LARGER

Bemitil first; a class-level thesis second

Our long-term thesis is a family of adaptation medicines guided by molecular and functional data. Actoprotectors and meteoadaptogenic compounds are the research frontier. A positive Bemitil result earns follow-on experiments rather than proving an entire class.

07 / COMMERCIAL FOLLOW-ON

Post-exertional recovery in ME/CFS

A disease-specific follow-on asks whether Bemitil biology can translate into improved post-exertional recovery. This requires separate disease evidence and a clinical program. The heat experiment is the lead; it cannot validate ME/CFS treatment. Existing chronic-fatigue patents shape the IP route.
